Under five years mr A underwent a transformation character-wise, he lost his inhibitions, made inappropriate sexual remarks and became the funniest man alive (according to him).
Though he never found other people's jokes funny. He was diagnosed with 'joke addiction'

His need to jest around was so great that he would wake up his wife in the middle of the night to deliver a punchline

The joker on the other hand suffered from uncontrolled fits of laughter, though a fictional character, this malady is known as the pseudobulbular effect resulting from brain damage (the specific kind of which wasnt revealed in the movie)
