The fact I even have to make a thread on this is mind boggling, but here we go. The five methods of sex determination and how they apply to three types of endosex trans people, we'll discuss a stretched version of them and the true version.
To start with we'll elaborate on what I mean when I discuss the five factors of sex determination. I won't link to the thread again so we'll make it brief. It has to do with five factors that can be determined from birth.
We'll start with a mid 20s trans woman who took HRT and has gone through surgeries to conform to her gender.

Karyotype as expected is 46 XY, SRY activated. This cannot be changed. So male without question.
External genitalia is modified, but at birth there was a penis. So this is another obvious case of male.

Gonads, once again functional testicles were present in the body before the orchi and they did descend in adolescence. So again.. Male.
Hormone profile, so even with taking hormones to suppress these levels it doesn't change that naturally their body ran rather typical and healthy male levels. So again, male.

Internal reproductive structures are clearly developed Wolffian, functional prostate present too. Male.
Karyotype - 46 XY SRY [M]
External Genitalia - Penis [M]
Gonads - Testicles, functional at birth [M]
Hormone Profile - Male typical levels would be there if not intervened [M]
Internal Reproductive Structures - Wolffian [M]
Conclusion: Biological Male.
Now stretch it.
Karyotype - 46 XY SRY [M]
External Genitalia - "Surgical Vagina" [N]
Gonads - No gonad tissue present [N]
Hormone Profile - No natural profile [N]
Internal Reproductive Structures - Partial Wolffian [M]

Still a biological male as null is just not factored.
Now for a trans man who is post-op, the rare one at that. We'll go 30s for this one.

Karyotype 46 XX [No SRY] [F] - Karyotype can't be changed, we know this and even if it could the blueprints are already written out.

External Genitalia - Vagina, Clitoris at birth. [F]
Gonads - Functional Ovaries that even produced eggs were there once upon a time. [F]

Hormone profile - The body had everything for a healthy female cycle. [F]

Internal Reproductive Organs: Mullerian structures were present at birth. [F]

Conclusion: Biological Female.
Stretching it now.
Karyotype: 46 XX No SRY [F]
Genitalia: Constructed pseudo-penis [N]
Gonads: Ovaries removed, no gonadal tissue [N]
Hormone Profile: No natural profile [N]
Internal Reproductive Structures: Mullerian Remnants [F]

Still biological female, again null not factored
A non binary person who is "DFAB." Early 20s, takes microdose T.

Karyotype-46 XX No SRY [F]
Genitalia- Vagina, clitoris [F]
Gonads- Ovaries. [F]
Hormone Profile: Natural female typical [F]
Internal Reproductive Structures: Mullerian [F]
Conclusion: Biologically Female.
There's nothing really to stretch, but okay.

Karyotype-46 XX No SRY[F]
Genitalia: Vagina, Clitoris.
Gonads-Ovaries [F]
Hormone Profile- Still a female cycle, even if suppressed by T.
Internal Reproductive Structures: Mullerian [F]

Without a doubt, biological female.
Even if you stretch the factors to be present applying, the closest a trans person can get is by replacing three of the factors with a "null" factor so if anything I guess maybe they can argue they artificially become a new sex? That's about it though. They don't change sex.
The five sex determination factors continue to hold strong even in the face of trans people altering their bodies. I suppose there is one argument that can made here but it only gives them one "opposite sex factor" which is hormone profile, still the same conclusion.
