Do not simply list your experiences and what you have done hitherto. Too many application letters are too vague.
The employer/project leader needs to know what you want to achieve and learn, and what you can bring to the group. Be specific.

Show your interest. Let your curiosity shine through. Tailor your letter for the announcement. For example, if you're applying for a marine ecology position, do not focus on your passion for diving and traveling. Focus on the topic at hand.

Think hard about what the research project is about. What questions are the group you'll be joining focusing on? How does the project align with your interests?

You do not need to have all the skills already. But it must be clear that you are really interested in the science and are eager to learn.
