Subaru's desperation really shines in this moment. Ever since the loss of Rem, Subaru's checkpoint moving forward with no regard for lost lives is a source of tension & anxiety. We've already seen through the post-Whale checkpoint that there is no set temporal rule they follow.
What really struck me was her hand shaking and her insistence on the word "human"— as if trying to put up a wall between him and her with it. She doesn't want anyone reaching out to her, shutting herself off (her ability itself is based on that idea/psyche)... or does she?
Can't go in depth here spoiler-free, but this scene offers another piece to the puzzle of Beatrice, one of the most intricately crafted characters yet. I'd point out the bits scattered in S1 to decipher this, but I feel it'll be more satisfying to the viewer to watch Arc4 unfold.
I like how his approach to Emilia is different at every restart. His tone this time is more impatient than understanding.
Subaru looks like he finally has a clear grip on things, thinking rationally and putting together information he has. But this isn't depicted as necessarily a positive; it's unnatural, as we see through Otto's perspective. Subaru's state of mind almost doesn't seem human.
We learn about another side of Garf's — while Frederica feels longing for him, in turn Garfiel has abandonment issues & feels betrayed. The layers & personal troubles of each character keep stacking up.
...I also like how, despite his attitude, Garfiel has a head on his shoulders with the way he deals with what he logically perceives as a threat. Much less brash than how a blue maid once acted, wouldn't you say?

What exactly is similar between Subaru and Roswaal at that moment? Why did Roswaal call him his partner in crime? Guess we'll find out.
Subaru is stuck all alone with nothing but his thoughts to accompany him—we see his mental state slipping as he ponders on Beatrice and all the carnage he witnessed and lived (well, died) through so far, desperately wanting to reset but unable to take his life.
And then comes Otto. Otto defies all expectations for the "trader" character archetype; we'd already seen his lack of ability/luck in his line of work as he was stuck with more oil than he could sell in Season 1, but this moment shows just how emotion-driven he is for a merchant.
And there comes my favorite scene. Subaru doesn't understand why Otto would help him; at this point, he's internalized the lessons he's learned from the candidates throughout Arc 3. Something as simple as someone lending him a helping hand feels odd to him. But there's more.
Video essays like "The Loneliness of Re:Zero" or "The Cruelty of the Respawn" go into this in detail; but in short, a recurring theme in RZ is the loneliness of Subaru as he goes through hellish timelines and tries to fix things himself. That's why this moment shakes him greatly.
After being betrayed by a perceived "friend", a new one steps in—and Subaru, used to both loneliness and distrust, doesn't take it in that easily, unconsciously twisting Otto's words into something that's easier to digest, more "realistic".
I really like that it's specified it's NOT because of the favors Subaru did for him, but simply because Otto viewed him as a friend—WANTED a friend. Subaru's world is flipped upside down, while we finally get a window into Otto's character & wants in this heartwarming moment.
This teary laugh is both happy and depressing. For a long time, he felt a disconnect from the world (even casually using words like "this time" or "restart" without anyone else understanding)— Otto snapped him out of that loneliness & brought him back down to the world.
